All right, as requested, here's my little demonstation/tutorial on how I use Adobe Illustrator - version 9 for me, on the PC. I also use a Wacom tablet, but it can still be done with a mouse. You just need a little more patience.

This tutorial assumes you have played around with illustrator a little bit so far, or at least have a manual on hand to do basic things like Rotate, Resize shapes.

The quick tools I use are Pencil, Rotate, Resize, Zoom, Selection and Lasso and finally the Pen tool set to subtract vector points (Do this by clicking on the pen tool hold it down and slide over to where it shows the minus sign. Good for removing excess on hand drawn shapes).

First thing is I always make at least 3 layers. To make a new layer look to the Layer's control box and click the button to the left of the trashcan. If you hover over it you can see it say "Create New Layer". For every click it will put in a new layer above the layer you have selected (the green one).

Even the most simple illustration you should think of it as foreground (layer 3) middleground (layer 2) background (layer 1). Whichever layer is closest to the top of the stack you see here is the closest to the surface, the first thing you will see.

Click the eye if you want to hide a layer. Click the blank box next to it if you want to lock it. And clicking again will reverse. Just this will make your work a lot easier.

At this point you should also decide how this piece is going to end up. If you want to print it, use CMYK for your colors. If it's just for web, have at it with RGB. I used CMYK - the color range isn't as wide, but it'll come out truer from screen to print than RGB.
