These images are from two very similar projects that I've just helped my friends with. They want to be able to hold bike frames on their alignment tables by the head tube. They both purchased used bench centers, and we had to make bull nose centers for them.

The fit up needed to be very close, so we were working to within a few tenths.

The other complication was that one of the centers on each set was spring loaded and utilized a rack and pinion to withdraw it... so we had to mill those rack gears.

Amino acid sequence alignment and phylogenetic tree of ginger CDPK1.(A) The amino acid sequence alignment of Zingiber CDPK1 [GenBank ID: KC544003] with Datura CDPK, DmCPK1 [GenBank ID: 163658596] and Arabidopsis CDPK, AtCPK30 [GenBank ID: 30699042]. Identical amino acids are indicated by asterisks, and similar amino acids are marked with single dots and colons. Catalytic domains (I–XI), Junction domain, and EF hand loops of CaM-LD domain of ZoCDPK1 are marked. The 15 invariant amino acid residues for eukaryotic Ser/Thr protein kinase were highlighted. Protein kinase ATP-binding site is shown by broken lines. Active site is shown in red box. (B) Phylogenetic analysis of ZoCDPK1 with all Arabidopsis CDPKs in the database. The tree was constructed using NJ method of Mega4 software. The numbers on branches showed bootstrap probabilities determined for 1000 re-samplings. The database accession numbers are indicated in parantheses after CDPK gene names.

It is necessary to form a stable dispersion of nanotubes in order to properly integrate them into polymeric systems. This can be achieved by treating them with acid to oxidise the tube surfaces. The tubes will then spontaneously disperse in an aqueous medium. The viscosity of these suspensions is analogous to that of polymers; it increases gradually with concentration up to a critical point (at about 0.7vol%) where entanglement occurs. A solid nanotube film has been formed by filtering the suspension through a 0.2mm membrane filter. Suspensions of relatively high concentration (>0.3vol%) yield films with random tube orientations but at lower concentrations (as in this sample), liquid crystal aggregation occurs and there is noticeable mutual alignment. The films exhibiting such alignment are tougher. This image was taken using a field emission gun scanning electron microscope (FEGSEM).

System

Carbon nanotube

Composition

Not specified

Reaction

Processing

A dilute (<0.3 vol%) suspension of nanotubes is filtered through a 0.2 mm membrane to form a solid film.

Applications

Carbon nanotubes may be considered a high performance mechanical polymer or an electrically conducting polymer but their greatest potential is in gas storage or as a filler in polymeric materials

Sample preparation

Solid nanotube films were prepared by filtration onto a 0.2 mm membrane filter under 0.6 bar negative pressure

Technique

Field emission gun scanning electron microscopy (FEGSEM)

It's a numbers game for me

Brace (for teeth) alignment tool i help make.

Forks need to be checked and aligned very soon after brazing. A straight edge and sight check allows me to asses a level plane across the axle line. Any post braze inaccuracy is quickly cold set and corrected.

It looked damn dangerous from the lens. I can imagine how difficult it would have been for the crane operator to align the column spot on.

Bridge over Deep Creek on the old highway one alignment into Gympie.

These ends are quite robust and don't have any misalignment issues throughout the build sequence. These particular end alignment tools belonged to the late Frank Herety. They're not pretty bits of engineering but do the trick just fine. Note the tiny breather holes on the inside of the seatstay. I will plug and fill these at a later stage.

The screen shows the light source alignment mode. After the dual light source is attached to the camera, the shutter is opened using the "B" setting and the user verifies visually that the sensors are centered in the film window.

Then the light source is moved until the bar graphs show equal amounts of light. When these levels are satisfactory, tests can be started.

Characterization and sequence analysis of GhMKK1.(A) Multiple amino acid sequence alignment of GhMKK1 (HQ828075) with AtMKK1 (NP_974619), NtMEK1 (CAC24705), LeMKK1 (AAU04433), and PcMKK2 (AAS21304). Identical and similar amino acids are shaded in black, and the docking domain is shaded in grey. The Roman numbers (I-XI) on the bottom indicate conserved subdomains, and a double-head arrow marks the active site motif. The serine and/or threonine residues in the conserved consensus motif, S/TXXXXXS/T, between subdomains VII and VIII of the MAPKKs are underlined, and the conserved consensus motif GXGXXG is boxed. The dots above the sequences represent activating sites, and asterisks indicate substrate specificity. (B) The phylogenetic relationships between GhMKK1 and other plant MAPKK proteins. The neighbor-joining phylogenetic tree was created using Clustal W and MEGA 4.0 software. The numbers above or below the branches indicate the bootstrap values (>50%) from 500 replicates. GhMKK1 is highlighted in grey. Each gene name is followed by its protein ID. The species of origin is indicated by the abbreviation before the gene names: Pc, Petroselinum crispum; Le, Lycopersicon esculentum; Gh, Gossypium hirsutum; At, Arabidopsis thaliana; Os, Oryza sativa; Zm, Zea mays; Nt, Nicotiana tabacum; Ms, Medicago sativa; and Nb, Nicotiana benthamiana. A, B, C, and D indicate the MAPKK group.

I thought my photostream was in need of a quick burst of abstract colour. This was part of a huge rusting circular drum on a disused railway line in Bristol, that was in the process of being restored and redocorated.

Before we strap a rocket onto it and send it screaming down the track we want to make sure the alignment is correct. The cars are hooked to a shuttle that is itself hooked to the sled. The shuttle ensures the cables are generating an even pull since the cars will not be in perfect sync and so the sled will be pulled straight down the track with no lateral force

Domain architecture and phylogenetic analysis of whitefly Argonaute2 protein.(A) Comparative domain architecture of whitefly Argonaute2 with other insects and C. elegans. Both PAZ and PIWI domains are present in all the analysed sequences. (B) Alignment of PAZ domain with other insects and C. elegans sequences. Figure shows high degree of homology between aligned sequences. Triangles indicate the presence of signature sequences responsible for efficient binding with siRNA [40]. Almost all the signature sequences are present in whitefly. (C) Phylogenetic analysis of whitefly Argonaute2 with other insects and C. elegans. Tree was constructed on the basis of full length sequence. Figure shows that the whitefly Argonaute2 clustered with N. lugens and aphids showing high homology between them. Similar result is observed in multiple sequence alignment (File S3).

I now do an initial check of the frame to check for alignment, make any adjustments that are necessary and plan my weld sequence.

Works well for now till I can put together a full table. Few small things to make for the bb tower.

Was drawn into this little nook in Florence by the architectural lighting in the alley... but turned around and pulled off what is probably my best example of timing and opportunity with composition thus far.

The fact he doesn't align perfectly with the lighting strip only annoys me slightly. ;-) But better the silhouette in the window... there was nothing I could have done to get both alignments perfect except hire a contractor to change something.

Of course these are all thoughts after the fact... in the moment one has no time for such analysis and it's just instinct and split-second reflexes. If I didn't fully appreciate this aspect before, after this photo I definitely do.
