A scaffold was shaped over a mold, attached to mesh, and seeded with stem cells. After 38 days in culture, the stem cells had grown to create a smooth, glistening surface. This process may lead to treatments for hip osteoarthritis that avoid the need for extensive hip replacement surgery.

Researchers have determined that the protein complex TFIID controls stem cell genes that repair skeletal muscle. This image shows human differentiated skeletal muscle fibers (myotubes, in green) expressing the protein MyoD (stained in red), which cooperates with TFIID in causing muscle stem cells to become muscle tissue. Cell nuclei are stained in blue. This discovery may help develop strategies that activate stem cells to repair muscle degenerated by aging or diseases like muscular dystrophy and cancer.

Induced pluripotent stem cells (cells from healthy skin that were “reprogrammed” to the embryonic state) were used as grafts on mouse skin with the blistering disease epidermolysis bullosa (EB). They repaired the EB-causing mutation in the collagen XVII gene and produced the functional protein (green), resulting in normal skin. These findings may help repair damaged skin in people with EB, preventing painful blisters.

Cartilage heals very slowly, due to a lack of blood vessels and other characteristics. One way to speed up natural cartilage repair and growth is to use tissue engineering, in which scaffolds, cells and other materials are combined to create functional replacement tissues. This image shows a three-dimensional biomaterial scaffold, consisting of multiple layers of resorbable fiber bundles that have been woven into a porous structure. The scaffold is infused with stem cells that grow to become new tissue as the fibers are resorbed. The fibers provide stiffness and strength in a manner that mimics healthy cartilage. This photo was selected as a 2012 winner of the BioArt competition of the Federation of American Societies for Experimental Biology (FASEB).

Immature muscle cells fuse together during development to form long muscle fibers with many nuclei. To identify factors involved in the fusion process, scientists studied fibroblasts—cells that don’t normally fuse. As shown in the microscopic image, adding a gene that makes a protein named myomerger to fibroblasts causes them to fuse together in flower-like clumps of fluorescently-stained cell nuclei. The protein works in tandem with another protein, called myomaker, to cause fusion. By gaining a better understanding of the processes involved in muscle development and regeneration, the research may help lead to new therapies for muscle disorders.

This microscopic image shows stem cells (stained green) migrating into a mouse skin wound to heal it. Cell nuclei are stained blue and the protein integrin alpha 5, a marker of migrating cells, is stained red. Researchers at The Rockefeller University have found that epithelial stem cells, which reside in a deep layer of the skin, “remember” wounds or other inflammatory events. This ability helps them respond faster to subsequent injuries.
