One of my best experiences I had in this trip was that I was able to see many peieces of notebooks written by Leonardo Da Vinci. The exhibition in the Uffizi was outstanding... See how he writes from right to left? There was a time (a long time ago) I wanted to try the way Leonardo did...it was so so hard...and I gave up within an hour! haha

This was the last time I looked at diatoms through a microscope - on 18 January 1983! It has relevance to the Fourier transforms discussed in the previous two posts. Notably the viewing of the back focal plane of the objective relates to the computed power spectrum.
The colour drawing on the second page shows the grating diffraction produced by the diatom pattern of apertures, the light source being the condenser illuminated ring which is part of the phase contrast setup.
The drawing on the 3rd page shows a similar diffraction pattern from an almost closed condenser diaphragm at the centre of the field: this analogue image corresponds approximately to the computed (digital) power spectrum.
This is a good demonstration of Abbe's theory of microscope resolution: the numerical aperture of the objective has to be great enough to capture the diffraction pattern of the image fine structure.
Mini notebooks with recycled children's artwork for the colourful pages.
