We used a novel transgenic mouse (TRAP line, Guenthner et al. 2013) that allows for the specific tagging of the neurons activated at a precise time. We made this mouse re-experience a specific memory and captured the neurons active at this very moment by making them express a fluorescent protein (tdTomato), so that we could image them later. We then made the brain fully transparent (CLARITY, Lee et al. 2016) and imaged its hippocampus by light sheet microscopy (Zeiss). Volume imaged: 2X2X2 mm. 3D processing and pseudo-coloring was done with Imaris.
The imaging of structures located deep inside the brain normally requires cutting it into very thin slices. However, with a revolutionary technique called CLARITY combined with light sheet microscopy, we can now make an intact brain fully transparent and image the whole tissue preserving its morphology. I used this trick to detect memory neurons throughout an area called dentate girus inside the mouse hippocampus. This part of the brain is the one creating new memories and storing them. With this revolutionary technique, we are now analyzing how neurons storing a specific memory connect to each other and how these connections change when memories are strengthened or attenuated. This will help us understand the neural mechanism of pathological forms of memory such as post-traumatic stress disorder, where traumatic memories are too strong, or Alzheimer’s disease, where memory is lost. ¦ Image#4_59

This is a bit of a damaged old print of my father's, but I rather like it. Taken at Yeovil Junction sometime in the early 60s, It shows ex-GWR 0-6-0PT 6435 (now preserved at Bodmin) and auto trailer W235W. Two groups of young spotters are on the platform and the loco crew are leaning out of the cab chatting to them. Quite an atmospheric scene.
Bit of a last minute decision to head northwards for this, but the evening clearance was from the north so thought it was worth a go... 57007 passes Brumber Bridge with 4M51 1610 Tees Dock to Ditton on 27th July 2012

I'm a bit nervous about the hotel I've booked. Due to a major medical convention in the city almost all hotel rooms have been booked, I've really struggled even with the help of the customer and the clout they have in the city.
But I did find somewhere... Le Nuvole Residenza d'Epoca will be my base for the 4 nights.
It is located in the historic centre of Genoa. This is a traffic free zone, so some walking from the Taxi to the hotel is required. A little tricky on the cobbled streets.
The lift in the corner takes me up to the 4th floor, where the hotel is located.
Everything in the historic centre is +500 years old. Note the heavily worn black marble stairs.

A bit of background on this photograph for those unfamiliar with Paraty (most readers, I assume). Paraty was built slightly under the sea level, and when there is a strong tide becomes a tropical Venice. When the sea retreats, there are plenty of water pool lefts to go around and make the most of your camera!

Un poco de información sobre esta foto para los que no conocen Paraty, que me imagino que deben ser la mayoría). Paraty fue construida ligeramente por debajo del nivel del mar y cuando hay una marea fuerte se convierte en una especie de Venecia tropical. Cuando la marea baja, quedan numerosos charcos de agua por todas partes para aprovechar la cámara al máximo.

Lobak - Malaysia Garden
The battered beancurd pastry skin was a bit oily.
The filling was a bit watered down with too much flour, but The streak of seaweed made it look more interesting.
Renee thought they tasted like 福建鸡卷 Hokkien-style Chicken Rolls.
