This series delves into the potentialities of architecture, design, and daily life, transformed by the limitless creativity and precision of AI. It captures a world where physical constraints persist, yet human ingenuity is amplified through the vast possibilities offered by technology. Protocol for Handling Collected Bees and Preparation
Introduction:
For taxonomic reasons and species conservation, it is often necessary to sacrifice a limited number of specimens from a population, similar to an ecological biopsy.
Bees are not as easily prepared as many other insects for proper identification.
Why?
Bees (Apoidea: Anthophila) are "vegetarian wasps" with voluminous scopae and hair structures for pollen collection. On the other hand, Crabronid wasps (e.g., digger wasps), the sister group of Apiformes, do not have such branched hairs. Bees, with their fluffy hairs, are therefore difficult to prepare so that the hairs are useful for diagnostic identification. Keeping specimens in alcohol and subsequent preparation with a needle usually results in damaging the fluffiness of the hairs. Thus, the fluffiness of bee hairs needs to be technically preserved for taxonomic collections and macro photography. This is not a trivial task and is technically more difficult than, for example, preparing wasps, beetles, or other insects. Here, two methods (A) and (B) for "hairstyling" bees for diagnostic purposes and photography are used:
(A)
After catching a bee alive, it is easiest to kill the insect in a killing jar with a small ball of toilet paper tissue soaked with a little amount of ethyl acetate. Preparation of the bee should be within a few days, while the insect is still relaxed. The advantage of this procedure is that the specimen retains its hair color and fluffiness. However, the disadvantage is that ethyl acetate is not well compatible with good DNA and RNA preservation for subsequent barcoding purposes. Alcohol preservation is preferred whenever specimens need to be kept for longer than a few days.
(B) The alternative protocol is more elaborate but preserves DNA well:
1. For scientific purposes, preserve a living bee in 90% EtOH, e.g., in a Falcon tube.
2. Remove the field label from the vial and save it.
3. Pour off the EtOH and specimen into a small dish.
4. Transfer the specimen with soft forceps into a Falcon tube containing hot soapy water detergent.
5. Shake and wash for at least one minute.
6. Pour the liquid and bee into a small kitchen sieve and rinse with hot tap water.
7. Transfer the specimen into a special drying glass with a net cover. The bottom of the glass contains toilet paper for absorbing fluid. Dry with a hairdryer until the hairs are fluffy.
8. Transfer the specimen into a moist two-chamber box (e.g., Sistema). Allan Walls uses a small Sistema box produced in New Zealand. The lower compartment contains toilet tissue dampened with soapy water to prevent mold. The upper, perforated container (with ~6mm drilled holes) carries the specimen.
9. Place the container into an incubator at ~50°C (I use a honey bucket warming cabinet).
10. Condensation may form on the cover of the moisture chamber. Avoid dripping it onto the specimen the next day. Remove the box carefully.
11. Place the specimen on paper secured with needles on a styrofoam pad. Pin the specimen with an insect needle or a minutia needle. Arrange legs, antennae, and wings with fixing pins.
12. Leave for at least three days to dry (depending on the size of the insect). Identify the species using taxonomic literature, DNA analysis, and AI-driven identification platforms such as iNaturalist, ObsIdentify, or others.

Protokoll zur Handhabung gesammelter Bienen und Präparation
Einführung:
Aus taxonomischen Gründen und zum Artenschutz ist es oft notwendig, eine begrenzte Anzahl von Exemplaren aus einer Population zu entnehmen, ähnlich einer ökologischen Biopsie.
Bienen sind nicht so einfach zu präparieren wie viele andere Insekten zur korrekten Identifizierung.
Warum?
Bienen (Apoidea: Anthophila) sind "vegetarische Wespen" mit voluminösen Scopae und Haarstrukturen zur Pollensammlung. Andererseits haben Crabronid-Wespen (z.B. Grabwespen), die Schwestergruppe der Apiformes, keine solchen verzweigten Haare. Bienen mit ihren flauschigen Haaren sind daher schwer so zu präparieren, dass die Haare für diagnostische Identifizierungen nützlich sind. Das Aufbewahren von Exemplaren in Alkohol und die anschließende Präparation mit einer Nadel führt in der Regel dazu, dass die Flauschigkeit der Haare beschädigt wird. Daher muss die Flauschigkeit der Bienenhaare technisch erhalten werden, sowohl für taxonomische Sammlungen als auch für die Makrofotografie. Dies ist keine triviale Aufgabe und technisch schwieriger als zum Beispiel die Präparation von Wespen, Käfern oder anderen Insekten. Hier werden zwei Methoden (A) und (B) zur "Frisierung" von Bienen für diagnostische Zwecke und Fotografie verwendet:
(A)
Nach dem Fang einer lebenden Biene ist es am einfachsten, das Insekt in einem Tötungsgefäß mit einem kleinen Ballen Toilettenpapier, der mit etwas Ethylacetat getränkt ist, zu töten. Die Präparation der Biene sollte innerhalb weniger Tage erfolgen, während das Insekt noch entspannt ist. Der Vorteil dieses Verfahrens besteht darin, dass das Exemplar seine Haarfarbe und Flauschigkeit behält. Der Nachteil ist jedoch, dass Ethylacetat nicht gut mit der Erhaltung von DNA und RNA für nachfolgende Barcoding-Zwecke kompatibel ist. Alkoholkonservierung wird bevorzugt, wann immer Exemplare länger als ein paar Tage aufbewahrt werden müssen.
(B) Das alternative Protokoll ist aufwendiger, bewahrt jedoch die DNA gut:
1. Zu wissenschaftlichen Zwecken wird eine lebende Biene in 90% EtOH konserviert, z.B. in einem Falcon-Röhrchen.
2. Entfernung der Feldetikett vom Röhrchen und Aufbewahrung.
3. Abgießen des EtOH und des Exemplares in eine kleine Schale.
4. Überführung des Exemplares mit weichen Pinzetten in ein Falcon-Röhrchen, das heißes Seifenwasser enthält.
5. Schütteln und waschen mindestens eine Minute lang.
6. Flüssigkeit und Biene in ein kleines Küchensieb gießen und anschließend mit heißem Leitungswasser spülen.
7. Überführen des Exemplares in ein spezielles Trocknungsglas mit einem Netzdeckel. Der Boden des Glases enthält Toilettenpapier zum Aufsaugen von Flüssigkeit. Trocknen mit einem Föhn-Haartrockner, bis die Haare flauschig sind.
8. Überführen des Exemplares in eine feuchte Zweikammerbox (z.B. sistema). Allan Walls verwendet eine kleine Sistema-Box, die in Neuseeland hergestellt wird. Das untere Fach enthält mit Seifenwasser angefeuchtetes Toilettenpapier, um Schimmel zu verhindern. Der obere, perforierte (mit Bohrer erzeugte ~6mm Löcher) Behälter trägt das Exemplar.
9. Pllatzieren des Behälters in einen Inkubator bei ~50°C. (Ich benutze einen Honig-Eimer Wärmeschrank).
10. Am Deckel der Feuchtigkeitskammer kann sich Kondenswasser bilden. Man vermeide, dass am nächsten Tag Wasser auf das Exemplar tropft. Entfernen Sie die Box vorsichtig.
11. Platzieren Sie das Exemplar auf Papier, das mit Nadeln auf einem Styropor-Pad befestigt ist. Nadeln des Exemplares mit einer Insektennadel oder einer Minutiennadel. Ordnen der Beine, Antennen und Flügel mit Fixiernadeln.
12. Mindestens drei Tage trocknen lassen (abhängig von der Größe des Insektes). Identifizieren Sie die Art mit Hilfe taxonomischer Literatur, DNA Analysen und KI-gesteuerten Identifikationsplattformen wie iNaturalist, ObsIdentify oder anderen.
Für die Makrofotografie wird in einigen Phasen ein Ultraschallbad verwendet, um Staubpartikel zu entfernen.

These two little buddies were photographed through a double glazed window, at an angle, with the wrong lens (a 70-300 about 2 meters away) and wrong settings (had to use whatever I had before they took off). Basic lighting adjustments were done but I also used Lightroom's incredible AI-driven removal of glass reflections plus some non-AI denoise.

Fountain of Happiness is an interactive installation that features seven sets of swings and light tubes designed to resemble fountains. Visitors are welcome to share moments of happiness on the swings and watch as the colours transform with each swinging motion. It invites both the young and young-at-heart to unwind, play and set off various lighting effects to infuse fun colours into the city skyline.
The installation leverages Alibaba Cloud's AI-driven sustainability solution, Energy Expert, to measure and analyse real-time energy consumption, creating a space where art, sustainability and technology could co-exist in harmony.
