This is the precipitation step of an RNA extraction protocol we've been using lately. Started from 5 grams of plant tissue, so there's a lot of precipitate. Next the whole things gets transferred into a kit column thing for further purification.

 

Will RNA be as popular as DNA? Doubtful.

Onderweg spotten we de Wild Ass (wilde ezel) en de nijlgau. De nijlgau of nilgai (Boselaphus tragocamelus) is een grote antilope uit India. 'Nilgai' is Hindi voor "blauw rund".

 

Aan het meer spotten we kraanvogels , flamingo's, pelikanen, ibis, spoonbill of lepelaar, ooievaar of blue bill, eenden of ducks ...

 

We ontmoeten de Koli vrouwen met bundels houten takken op het hoofd.

 

Description(Physical Details): Set of tools for dental extraction and other basic dental surgery in velvet-lined wooden box. Box has brass corners guards and braces at corners, and a locking front. The top tray contains 12 instruments with ebony handles, and 12 with ivory handles, including 15 excavators of varying sizes and shapes, one universal handle, and 7 scalers. The top tray fits the instruments with flat-laying openings, a sloping area in the middle on top of a mirrored base, and have a gold-print filligree as a border around the outside. The bottom tray has two imprinted velvet areas, and includes 7 extraction forceps, 3 stump extractors with ivory handles, and 2 stump extractors with ebony handles.

A CH-47 Chinook crew from Bravo Company, 1-126th General Support Aviation Battalion, was called to assist aviators from the Naval Strike and Air Warfare Center from Fallon, Nev., on Jan. 5, 2012. One of their Navy MH-60S Seahawk's had crash landed during a training mission high in the Toiyabe National Forest near the U.S. Marine Corps Mountain Warfare Training Center (MWTC), northwest of Bridgeport, Calif. All four crewmen aboard were able to walk away from the Dec. 21 incident but now the Navy wanted their aircraft back. The California Army National Guard flight crew from Stockton landed on a bald hilltop at 9,200 feet and was escorted to the crash site by Marines from the MWTC. Over the next few hours U.S. Marine rigging specialists from the Combat Logistics Battalion 17, specially called up from Camp Pendleton, worked with their fellow Devil Dogs trained in alpine, rock, and snow warfare, to prepare the pre-stripped helicopter for extraction. Taking guidance from a Naval MH-60S crew chief and the experts flying the tandem rotor hauling machine, the multi-service recovery team was able to pluck the 15,000 pound fallen bird, which was painted as an opposition force "enemy" aircraft, above the pine trees, down the canyon and back to the MWTC. Everyone watching, including representatives from the U.S. Forestry Service and the MWTC Environmental Office applauded the safe and successful operation. The aircraft, which lost its tail end during the hard landing, will rest on the Marines tarmac while it is subjected to further pokes and prods as key personnel continue to investigate how the incident happened. SES rescue exercise.

 

You are an SES emergency worker. You arrive onsite in your rescue vehicle. You know that a bus has been in an accident. It is pitch black.

You can't see the bus, the fires, the damage. You don't know how many victims there are and what their ailments are.

 

The team kicks into gear. Cones go out marking the area whilst the generators are setup and emergency lighting comes online.

 

The first wave locate the bus, locate the fire hotspots, assess the safety and then cautiously enter the bus.

They move through the bus, taking note of the doors that will not stay open, the blown airbag, the debris.

They locate the driver, a man in the first section with missing limbs, a man dangling from the roof emergency exit, the pregnant lady and the impaled person.

They reassure everyone that they are there to help and form a plan.

 

The second wave has news from the first wave, directing them to those that need help, in what order and what to watch out for.

Their plan is to maximize survivors.

 

They wedge the main doors open and begin.

 

The driver is removed on a back brace. The man with missing limbs is carefully moved through the bus aisle and out the door. The 8 month pregnant lady is in a lot of pain and shouting.

She is carefully monuvered out of the door but being 8 months pregnant is a challenge. The 63 pound man hanging through the hatch is lowered and rescued.

 

Meanwhile the injured are in triage and being looked at by a doctor not far from the scene. The man in charge is constantly assessing the situation and dangers.

Finally they need to cut free the impaled person. Then on a back brace,remove her from the bus.

 

All of this whilst the patients are in pain, yelling and screaming.

 

I performed the part of an annoying media person whom was keen to get photos but needed to be kept in check and monitored to make sure I also did not end up hurt.

 

This was only a scenario. It was play acting.Having said that, the exercise was very real. Their actions were very real. People needed to be extracted and the SES team had to solve problems and overcome.

They did and I was very impressed.

  

This was performed in a Mercedes Benz O305G 587 articulated bus on the SES Training Course.

Leaves of Hibiscus rosa-sinensis are processed using different methods depending on the intended application. Using three different processing methods, we investigated the effects of processing on the proximate constitution of the leaf. Result demonstrated that the fresh raw leaf had moisture content of 82.30 ± 0.42%, which were significantly (p<0.05) reduced by drying but not extraction and blanching. The protein content of the raw leaf was low (1.80 ± 0.10%). Extraction and blanching reduced the protein content, whereas drying increased the protein content significantly (p < 0.05) for raw dried leaf powder and blanched leaf products. The raw leaf contained vitamins A, B2, C and E, which were significantly reduced by extraction and blanching, but were concentrated by drying. Anti-nutrient contents of the raw leaf were low and were reduced to negligible levels by the processing techniques employed. Comparing the nutrient and chemical constituents with recommended dietary allowance (RDA) values, we found that the leaves contain an appreciable amount of nutrients, minerals, vitamins, proteins and phytochemicals and low degree of toxicants. These findings suggested that the treatment method employed in processing this leaf affected the proximate composition, and this should be considered in utilization of this leaf (and other leaves) product in various food and pharmaceutical formulations. Various heat processing techniques applied during the preparation of the processed products from Hibiscus rosa-sinensis leaves, caused adverse effects on the chemical composition of the processed leaf products. This was evident especially for the vitamins and minerals constitution of the processed products. More so, blanching and drying caused a significant reduction in the nutrients and anti-nutrient composition of the formulated samples. While the best processed samples were the dried powdered products, especially the RDLP, whereas the worst processed samples were the extracts, notably B2LE. It is recommended that other processing techniques such as freezing, solar and spray drying and ethanol extraction can also be applied in order to determine their effects on nutrient retention and anti-nutrient reduction on the plant leaves and compare it with the results of this study.
