La voie tire son nom de l'ancien théâtre Molière, qui se trouvait à proximité, lui-même portant le nom du comédien Molière. Il s'est également appelé « passage des Sans-Culottes » sous la Révolution et « passage des Nourrices ».
Le passage est ouvert vers 1791. Il est inscrit au titre des monuments historiques en 1984
Il fait l'objet d'une rénovation lourde entre 2017 et 2022, accueillant désormais 34 logements sociaux et 9 commerces, parmi lesquels une papeterie et un bar à vins.
