We have recently established a novel immunohistochemistry-based procedure to detect expression of up to 5 proteins in formalin-fixed paraffin-embedded (FFPE) tissues and cell samples. The procedure is based upon sequential combination of horseradish peroxidase enzyme labelling that is visualised using the diaminobenzidine (DAB) brown or yellow substrates with the alkaline phosphatase enzyme labelling revealed using chromogens of different colours (i.e. Fast Blue, Fast Red, Green).

 

The present five colours immunostaining of a single section of human tonsil shows expression of CD34 (yellow) in vascular endothelium, CD138 (green) in plasma cells and epithelial cells of tonsillar criptae, CD3 (purple-red) in T-cells, the B-cell associated transcription factor PAX-5 (nuclear, brown) in germinal center B-cells and CD21 (blue) in follicular dendritic cells within a germinal center. Note the absence of non-specific background labelling in the sections and no nuclear hematoxylin counterstain was applied.

 

The immunostained slide was scanned using the NanoZoomer Digital Pathology System (Hamamatsu). The authors thank Mr D Oukrif for his technical support using the NanoZoomer.

Mass spectrometry imaging (MSI) is a powerful analytical technique with which one can acquire a map of chemicals in biological samples. Combining this technique with traditional histology, which is a type of tissue-staining procedure done in medical labs to visualize tissue components, can provide complementary information by linking the tissues’ structure to its chemical makeup. However, improving the accuracy of image overlays is crucial for better image clarity down to the single-cell level, which is essential for digital pathology. Our photo is a mouse brain tissue collected on a glass slide that was laser-etched with fiducial markers (crosses), analyzed using MSI (one-third of whole tissue) and stained with hematoxylin and eosin dye post-MSI. Results indicated that the fiducial markers provided significantly better alignment and accuracy. This research demonstrates the feasibility and benefits of integrating MSI with optical imaging, marking progress towards improved diagnostic capabilities in digital pathology.
