In 2015, Hospital Epidemiologist Dr. Tara Palmore ‘dons’ personal protective equipment (PPE) in the Special Clinical Studies Unit (SCSU). The anti-cancer drug pembrolizumab has shown promise in slowing or stopping the progression of progressive multifocal leukoencephalopathy (PML), a typically fatal infection of the brain caused by the JC virus (JCV). This finding comes from a small-scale study by scientists at National Institute of Neurological Disorders and Stroke (NINDS), part of the National Institutes of Health.

The Special Clinical Studies Unit (SCSU) is an inpatient unit at the NIH Clinical Center in Bethesda, MD., designed with state-of-the-art infrastructure that allows for isolation capabilities and infection control while patients participate in clinical research studies. The unit has been in use since the summer of 2010.

Hanna Kim, M.D., M.S., is a Lawrence Shulman scholar in the NIAMS Pediatric Translational Research Branch. Her research seeks to ascertain the pathogenesis (the biological mechanisms leading to a disease) for juvenile dermatomyositis, an inflammatory muscle disease. The results of these studies may lead to new biomarkers and potential treatments for the disease. In this photo, she is examining a pediatric patient in the NIH Clinical Center.

Dr. Denise Ney, a researcher in nutritional sciences, follows over 160 patients with phenylketonuria (PKU). PKU is a genetic disorder caused by deficiency of an enzyme that converts the essential amino acid phenylalanine to tyrosine. To prevent brain damage and cognitive impairment those with PKU must follow a lifelong, low-phenylalanine diet through restriction of their diet and the use of a bitter tasting amino acid formula. Dr. Ney provides education on the importance of nutritional management of PKU, and under the auspices of the Orphan Grants program is conducting clinical studies to establish the acceptability and safety of unique foods made with whey protein for PKU.

Abstract: Magnesium is vital for many biological processes such as energy metabolism, nucleic acid, and protein synthesis, signal transduction, and cell division. Impairment of magnesium homeostasis is associated with many diseases, especially cardiovascular diseases, hypertension, type 2 diabetes, and cancer. More than 300 million people worldwide struggle with type 2 diabetes, and this number is growing exponentially. Clinical studies have shown that serum magnesium levels were decreased in patients with type 2 diabetes and that magnesium supplementation has positive effects on glucose metabolism. In this study, glucose consumption and the expression levels of glucose transporters (ght1, ght2, ght5) were investigated in magnesium transportrestricted mutant strain of Schizosaccharomyces pombe yeast, which is similar to mammalian cells in terms of biological processes and genetic mechanisms. In mutant strain with limited magnesium transport, glucose consumption increased due to the increase in magnesium added to the medium. The expression level of ght1, ght2, one of the glucose transporters, increased in 30 mM Mg2+ supplemented medium, decreased at higher magnesium concentration (75 mM), where optimum growth was observed for the strain, and there was no significant change in the expression level of ght5. Our findings indicate that the glucose transporters ght1 and ght2 are regulated by a different mechanism than ght5.
