En un tubo con anticoagulante (EDTA o ACD), se colecta sangre total periférica y se centrifuga por cinco minutos a 3000 RPM (radio del rotor: 15 cm). De esta manera, obtendremos una capa blancuzca sobre el paquete de células rojas y debajo del plasma.

Tras retirar el "buffy coat", el tubo debe contener sólo la fracción eritrocitaria y la capa superior de plasma, como se muestra en la fotografía.

En un tubo al que previamente se le ha añadido disolución de lisis con detergente no iónico, colocar el "buffy coat" obtenido.

Con ayuda de una pipeta de transferencia o micropipeta, retirar la capa blancuzca con la menor cantidad posible de plasma o paquete eritrocitario.

The red part is erytrocytes (red blood cells), and the yellow part is plasma. In the middle there is a small part of byffy coat; White blood cells and platelets.

This image shows the buffy coat layer between the red cells at the bottom and the yellow plasma. The buffy coat is a highly concentrated area of white cells. Sometimes buffy coat slides need to be prepared for patients with extremely low white cell counts.
