La versión Murciélago LP640 se presentó por primera vez en el Salón del Automóvil de Ginebra en marzo de 2006, como un adelanto de la versión cupé. Es una versión renovada del Murciélago, equipada con un motor de 6,5 L que produce 640 CV mejorando sustancialmente el rendimiento. Al igual que en el Murciélago, el motor está montado en la parte trasera, con la transmisión en la parte delantera del motor y el diferencial detrás de ella, en lugar de un transeje normalmente visto en coches con motor central.
